I think the most important reason of 6 hits with F is boss up and down, and the opportunity of shoot.
In x2poet's run, he made the boss in the left, but most of F only hit 5 times. He shot in the bad moment.
Maybe you can jump and change the shoot of opportuniy (y axis).

Sorry, I didn't work it after last upload.
After compare with the J and U version, I found a very big trouble. The reason of U version be faster than J version when Jack in is the less lag frames. Sometime U version's dialogue is longer than J version's, but both of them use the same RNG(RNG stop work when lag frames). It's mains that if I can't find the enough way to improve, I would miss the best luck, specially when can only use the last folder.
The nico run's quality is bery very high, and very hard to be improved. In this 10 minutes wip, I only saved very few frames, and saved fewer RNG, because I shoot the more times in teaching bittle, and cost more RNG.
So I have to stop now.

All saved 1846 frames, now!
Part1, I lost 18 frames, because I only use shotgun to hit helicopters, but it can save 60 frames in boss battle.
Part2, saved 115 frames. I give up get a card which add the grenade's power. This card add very low power to grenade, 100 to 120. x2poet got this card cost about 200 frames. But I mustn't use any shotgun or grenades. So I have to use knife to kill soldiers, and got the very large lag frames.
Part3, saved 580 frames. Manipulate luck to reduce lag, and didn't take grenades. Taking these grendes would cost near 500 frames.
Boss fighter, lost 94 frames, because I have the less grenades. 26 shotgun(200) + 20 grenades(100) = 7200 the boss HP!

Mission3!
Totle Improved 1263 frames!
Part1, avoid some foes appear.
Part2, manipulate luck and kill some bats. Which bats should be killed is all luck.
Part3, manipulate luck to make dive_soldiers appear early.
Boss, I found a faster way to use metalslug's Vulcan. I can shoot 1 more time in every 10 shots. So I made the boss being mad quickly. But I didn't always use it. Becuse the more bullets in screen the more lag.
I choose the minimum distance to shot, and make the less bullets in screen. So I got the less lag.
When I shot the last cannon, I thrown the metalslug atonce. If no slug in screen, the scene fade quickly. I found it in mission2's boss fighter.
So though my bombs are 1 less than the published run in boss fighter, I still saved 130 frames.

Mission2 completed! I have improved 806 frames.
I found the way to skip the first fighter helicopter(650HP). First, I can't save a prisoner to get H. 2nd, wait for blockhouse fire. I think the reason is RAM. If I save the prisoner, his memory address would be released, and can't skip helicopter. When the blockhouse firing, there're not enough memory to put helicopter.
In mission2 part2, I left 5 firebottles(80 power) after beat submarine(600HP). It haven't costed any more frames. Because if I can't skip next 2 helicopters, I would use these 5 firebottles to hit them. But luckily, I succeeded to skip them.
Boss fight. When emptying the Rocket Launcher in the air, I can't use handshot atonce, so I throw one firebottle. I must hit 3 soldiers first, the yellow soldier would appear. I found a best way to save Laser's arms. 10 frames, 32 damage and 0.5 arms! Yes! 0.5 arms.

This is my wip. I have completed mission 1 and improved the publish one 121 frames.
In mission 2, I plan to give up metalslug(because the slug is very slow), and take H to the end to manipulate luck L.
But this game's manipulation luck is like the hell. I'm trying skipping the first helicopter of misssion2.

I found a faster way to use H. zk547, you can use it in mission5's 2nd boss.

New improvment
I forgot to use reset in the title, and improved another 2 frames at begain. Totle saved 77 frames.
Mothrayas, replace the movie file, please. I have tested that this file sync perfectly, and have Modifyed the information, too. The info's "frame" have been reduced 100 frames, the "time" has been reduced 2 seconds. But I think no need to replace screenshot.
I hope no one has encoded yet. This is last, I don't plan to improve again.

Why you let boss go to left? Because of the less death-ray in screen and the less lag?
And what's meaning "3 frames" ? why not 2 or 4 frames? Because metalslug is 30 fps game in 60 fps system, every 2 frames have 1 frames which no effect.

Sorry, I made some mistake. Laser gun shot 4 times when pass A, not 5 times. So Laser's real arms is equal to 250, and 4000 damage in all!
After my test, I think you can choose the fastest way to shot. If you can keep every flame shot's main flame hit boss 6 times(when jumping, you can hit 6 times, too), only need 31 or 32 Flame shot. You have 1000 frames to use L, so in these 1000 frames, try to use most F. F every 32 frames 1 shot, in normal place(if no lag). I think you have enough time to use F, 1000/32+1≈32 (1 is first shoot).
Edit: The middle's Y pixel is higher than side, maybe in the middle is faster than in the side, when using F.

Jump and shot upward, I don't think shot sideways can damage mosquito. In the first 26 frames, F has the big power. After 26 frames, only side flames working.
But after carefully watching, I think you no need to redo it, because your F's main flame has hit mosquito 20 times, it's enough. It's seemed F has the large hitbox. You can go on doing next boss4, and test how F hitting this boss has the max power. I think only shotting upward on the ground is enough.

Tested, 2player can't get anything, but you can throw them when you meet first boss, Mr Allen, and manipulate luck for bombs from motorcycle-soldier.

Do you know how the FlameShot work? FlameShot include main flame and side flame. Main flame power is 20 and hit 13 times in 26 frames, if no lag. The side flame power is from 0 to 5, but hit more long time. F's all power is 314. The data's change you can see: http://tieba.baidu.com/p/1634986592
So you should jump and use F to shot mosquitoes.
In the beginning of mission5, you can try to throw the plane at first time. I know in 1player, there is a chance to get F.

Very high quality play and awsome glitches be used.
I think you can improve the 2 players, too.
2players run will cost lots of frames in slot machine.

Do you mean pack up C will cost more 100~120 frames, or you have tested that use C is slower than H?
These mosquitoes' HP are 140(I'm not sure every mosquitoe has the same HP). Don't forget C's smoke can damage mosquitoe, too. But all the C and H are slower than F, because F can hit more mosquitoes in once attack. I don't konw C or H is better, but you can manipulate luck and use F to hit most mosquitoes.

This nico's run is 100%, and he use the J rom. But he can't get the 8th big diamond, the 8th diamond is not very necessary.
