Only if the game makes something happen for each button press and/or is programmed exceedingly poorly (I have never seen any console or handheld game lag from mere button presses). It can't hurt to check that it's not slower, though.
Legend of Zelda: aLttP lags on too many button presses. I discovered this on my latest WIP. I began 4 frames slow than flag because of this.
Oh, and I think I lost a few frames on Rockman World 4 by holding down buttons too long. So it's probably good to go for minimal button presses.

Yeah, and if the poles get in your way, you have the few seconds of Mario grabbing them and then jumping off them.

What does a 100% run include, though? Do you have to win the cow? Do you have to bomb open every secret hole? Do you have to open every treasure chest in the game, no matter how worthless? Do you have to get perfect records on every mini-game?

I could check, but I thought that getting the star just left you where you were (Castle Secret Star), and swimming out took you outside the castle. Except what happens when you die in the Metal Cap course? Oh yeah, and has anyone thought of more ways to abuse the "exit course from pause screen" option to save time?
