Total improvement in in-game time: 170 frames, or 2 seconds and 50 frames.
The times given are the in-game timer values. The value after the final colon pair indicates the in-game frame counter, and is restricted to the 0-59 range. It is updated every frame (except during lag frames), and increments the in-game second when resetting to zero.

Green Hill Zone 1
An obvious slope jump, using flight to avoid breaking the monitor, combine to a large speed boost. Improved precision until the end carry the day.
Green Hill Zone 2 & 3
More precise play in the beginning of the acts. That is it.
Spring Yard Zone 1
The most obvious improvement -- who needs speed shoes when you can abuse the enforced screen boundaries of flight to get nearly the same speed?
Other Comments
Possible improvements: If one were to accept damage in the run, Scrap Brain 2 might be improvable by 6 frames by getting hurt at the saw. There are no other improvements of which I am aware of at this time, and all my attempts at improving the other levels failed.
