I added a ton of notes in one of my previous posts and I figured out a good route. And I don't have to forfeit part of my WIP if I accept a ~1 second loss, which is negligible. I should be making good progress soon

This is one reason why I'm going to wait quite a while after my TWW TAS is finished before submitting, because I know every little encoder is going to want to make their own encode for their channel, and then TASVideosChannel is just going to upload it and get all the views. TBH, it's a load of bullshit. But I suppose they won't make an exception for anyone, for the sake of "organization".

Pirohiko and I are working on a visualisation to display the brothers' stats, their abilities and current selected action icon IDs, the # of uses of bros attacks (until they learn Advanced versions), Enemy stats, items, coins, timer, along with the TAS. The video is going to be published in Youtube and nicovideo.

I know the TAS is already finished, but this might be useful for speedruns in the future; if you only land the first action command of Thunder Bros., the DEF-Down status reduces enemy DEF to 0 rather than 1/3 of its value.

The TAS never gets Thunderhand. Fights take a few minutes but getting the Hand probably takes 2 to 3 minutes. Hardly imaginable that it saves time..
E/ themushroomkingdom says Hammerbro has 130 DEF and Boo has 1000 DEF. In your video it looks like you assume both had the same DEF. Someone should look into the memory to see how the enemies' DEF behaves during battle.

Yes, I'm the same person, and I didn't know you could skip Thunderhand entirely. For an all-bosses run (I don't know how you'd define "100%" in M&L), it might speed things up a bit, though.
As for how it affects enemy stats, here are the exact multipliers for all button combos:
B - 0x00 (0%) DEF single
BB - 0x1E (12%) DEF all
BBB - 0x55 (33%) DEF all
Adv BA - 0x00 (0%) DEF single
Adv BAA - 0x46 (27%) DEF single
Adv BAAB - 0x48 (28%) POW single
NOTE: As far as I know, if an enemy is "immune" to attacks, like Glurp to Jump, Troopeas to Hammers, Jojora to anything, etc., this won't affect the damage they take. A way to test the difference is to use Mush Badges; if you only do 1 damage with a bunch of Mushrooms, then the enemy's immune to a certain attack.
I won't get into the minefield of a 100% definition(yet); but I do hope a run along those lines can be done. The any% becoming a demo for a few major skips was a huge disappointment for me, (as interesting as those glitches are) as, for lack of a better term, it looked like the any% was going to able show the whole game off.

But if it doesn't do those glitches, it will be judged as a playaround, since it doesn't complete the game at the fastest time. I don't think the run would be interesting enough to skip the glitches though.

Technically, an all-equips run is impossible; you can only get either Piranha Swing or Piranha Suit from the Piranha Bean boss. That and it wouldn't be very interesting, due to you having to get Swing Bros. Advance and fight a bunch of random enemies to get all of the equipment (and if including the pins, getting 35+ of all beans). I think no-sequence-breaking (or at least all bosses fought in proper order) should be about the maximum requirement.
All that said, I would eventually like to see a "more completed" run at some point in the future.
"No sequence breaking" is a legit category, much as it's causing me problems to get adelikat to admit its existence. There's already the occasional run in that category on the site already.
No sequence breaking is good if you define it rigorously - do you have to see all plot advancing cutscenes and fight all plot advancing bosses in order, for example? Or do you have to as well solve every puzzle that a casual playthrough would need to to advance? Etc

Could you use swing bros advance to steal Piranha Swing, then game over and fight again for Piranha suit?
